The importance of understanding how matrix stiffness affects cell behavior cannot be overstated. In a recent study, researchers investigated the relationship between matrix stiffness and hepatocellular carcinoma (HCC) cell proliferation.

The study found that increasing matrix stiffness led to an increase in HCC cell proliferation. This was associated with stiffness-dependent regulation of basal and hepatocyte growth factor (HGF)-stimulated mitogenic signaling through extracellular regulated kinase (ERK), protein kinase B (PKB/Akt), and signal transducer and activator of transcription 3 (STAT3).

To investigate this further, the researchers used two different cell lines - Huh7 and HepG2 - which were cultured on soft or stiff collagen-I-coated polyacrylamide gel supports. The results showed that the proliferative index of both cell lines was significantly higher when they were cultured on stiff supports compared to soft ones.

In addition, the researchers used siRNA to interfere with β1-integrin and focal adhesion kinase (FAK) expression in HCC cells. This inhibition was found to prevent stiffness-dependent proliferation in these cells.

The study also investigated how matrix stiffness affected cellular spreading in HCC cells. The results showed that stiff supports led to increased spreading of both cell lines, which was associated with an increase in the surface area of individual cells.

Overall, this study highlights the importance of considering the role of matrix stiffness in understanding the behavior of HCC cells and its potential implications for cancer development and progression.
